Mark Watkin (マーク・ワトキン, Māku Watokin) is the champion of the rural boxing club. Jonathan is meant to fight him as a challenger, but Dio Brando volunteers to have a match against Jonathan instead of Mark. The referee and Mark accept, and Dio emerges victorious.

Winzaleo the Lion King (獅子王ウィンザレオ, Shishiō Winzareo), Eijkman the Lightning Knight (イナズマの騎士アイクマン, Inazuma no Kishi Aikuman), and Caineghis the One-Eyed Man (独眼のカイネギス, Dokugan no Kainegisu) are three of the only five knights to survive the trial of the 77 Rings, winning the challenge over a century before Tarkus and Bruford in 1563. Winzaleo won the trial in 1327, Eijkman won in 1389, and Caineghis won in 1408.

Winzaleo carries a giant battle-axe, Eijkman utilizes an electrified halberd, and Caineghis wields a poisonous scimitar. Their appearances are changed dramatically from the manga.

Winzaleo is described in the game's Profile Mode as a knight who earned the title of "Lion King" due to his fierce appearance, skill with axes, strong build, and chivalrous spirit.

Father Styx (スティクス神父, Sutikusu-shinpu) is an alcoholic priest sent to Mexico via the boat Jonathan and Erina board in order to convert the locals. During the voyage, the unhappy priest drops his cross, which falls through a grate and lands near Dio's coffin. While retrieving the cross, the curious priest triggers a booby trap, causing a metal ball to shoot out and obliterate his face.
