I feel like the idea that the Stone Mask can help you unlock a stand is meant to mean that as a Vampire, you would be stronger than normal humans and much more likely to be able to gain a Stand. It's not really a direct way of gaining a stand, more so a way to evolve the human body, like a stand is.
